Anotation | Spectral flow cytometry is a highly innovative technique that allows simultaneous measurement of higher number of fluorescent markers in contrast to conventional flow cytometry. With the newly available spectral cytometer Aurora (Cytek) in our laboratory, it is a challenge to design and optimize a new panel for unique but very rare extramedullary myeloma samples. Extramedullary disease in multiple myeloma (EMD) represents a highly aggressive stage of multiple myeloma (MM) characterized by pathological plasma cells evasion of the bone marrow microenvironment and infiltration of soft tissues and organs. Patients suffering from EMD have an unfavorable prognosis with overall survival of up to several months and are typically resistant to standard treatment. Therefore, more detailed research on extramedullary myeloma has a great potential. The aim of the proposed project is to design a spectral flow cytometry panel for the characterization of individual immune cell subtypes and for the detection of expression of therapeutic molecules in extramedullary myeloma samples. The newly designed panel will allow us to use two multi-color tubes for spectral flow cytometry, compared to the seven eight-color tubes from the original panel in the SGS10/LF/2022 project which the current project is based on. The main advantages of the new panel will be saving the amount of sample needed for analysis, reducing the time of analysis and decreasing financial costs. |
